Probing Two-pore channel 2 in LRRK2-Parkinson’s in vivo
Parkinson’s disease is a major cause of disability in the elderly, the second most common neurodegenerative disorder above 60 years of age. Mutations in a large gene, LRRK2, have been linked to Parkinson’s. In a number of model organisms, expression of LRRK2 mutations recapitulates the loss of dopaminergic neurons and consequent movement deficits. This project focuses on the effects of LRRK2 in the genetically amenable organism, Drosophila. In particular, it focuses on the role of LRRK2 at the lysosome, the recycling organelle. We have a number of new genetic manipulations of the two-pore channel and novel compounds that may provide a disease-modifying therapy. The appointee will work in a lab, with other technicians, 2 research students and a post-doc all working on a range of neurodegenerative problems. This project is funded by a grant to Prof Sandip Patel (UCL), Dr Chris Elliott and Dr Sean Sweeney (York) from Parkinson’s UK.
Role
To provide technical and scientific support to research project in the laboratory. This will involve maintenance of Drosophila stocks, and characterisation of the novel mutations
Maintain accurate records of work undertaken, including reports, using appropriate (bespoke) software
Liaise with appropriate staff and external service providers to ensure the work area and equipment are kept operational
Skills, Experience & Qualifications needed
HNC, HND, Degree or equivalent in appropriate science discipline or appropriate experience
Relevant theoretical knowledge in Biology
In depth knowledge of relevant safety regulations and procedures for example, COSHH and risk assessment
Knowledge and ability to implement practical techniques used in relevant work area
Evidence of having contributed to activities using a variety of sometimes specialist technical skills
Experience of Molecular Biology (e.g. PCR, Western Blotting); training in fly work is not assumed and can be provided but will be advantageous
Experience of working on own initiative and as part of a team
This is a fixed term contract for 18 months.
Interview date: 30 June 2022
